InVivoKines™ – Proteins with Enhanced Activity & Stability
InVivoKines™ are a new generation of recombinant fusion proteins for immunotherapeutic, preclinical and translational in vivo research, developed in-house by AdipoGen Life Sciences. InVivoKines™ are Fc-based fusion proteins using the Knobs-into-holes (KIH) technology.
- The KIH-Fc technology allows Fc heterodimerisation to create a structure with two different arms, to design naturally occurring active monomeric, heterodimeric or homodimeric proteins.
- The KIH-Fc domain enhances the plasma half-life of the cytokine which substantially improves the pharmacokinetics (PK) while maintaining activity.
- The KIH-Fc domain engages with the Fcγ receptors (FcγRs) and the complement C1q, which may result in ADCC, ADCP and CDC.
- The KIH-Fc LALA-PG mutations inhibit binding to FcγRs and C1q while FcRn binding, and Fc stability remain unaffected.

Functional Grade Antibodies – FuncAbs™
Antibodies displaying an agonist or antagonist activity (functional grade antibodies (FuncAbs™)) are powerful tools for mimicking or blocking physiological functions in vitro and in vivo. Functional grade antibodies are available free of preservatives and tested for low endotoxin content and may be used for activation, neutralising or blocking studies, both in vitro or in vivo.

Recombinant Monoclonal Antibodies – RecMAbs™ – Produced with No Use of Animals
To promote the advancement of non-animal alternative approaches (3Rs), AdipoGen implemented the antibody phage display technology in-house to develop monoclonal antibodies without the use of animals.
Using this technology, AdipoGen was a forerunner in releasing the first recombinant monoclonal antibody for research on the market in 2010: anti-APRIL (mouse), mAb (rec.) (blocking) (Apry-1-1).
Since then, their portfolio of fully validated recombinant antibodies made with the phage display technology continues to grow, with a focus on functional antibodies that can block or activate their respective target.
Key Features of AdipoGen's RecMAbs™:
- Developed from a human antibody phage display library and produced in mammalian cells (CHO or HEK 293).
- Show similar properties compared to monoclonal antibodies developed in mice/rat/rabbit (e.g. affinity in the low nanomolar range).
- Ideal for conserved antigens (which are poorly immunogenic in animals) to detect conformational epitopes (e.g. GTP-bound proteins) or protein modifications (e.g. phosphorylations, ubiquitinations).

Small Molecules
AdipoGen Life Sciences' business division “Small Molecules/Biochemicals” supports the overall product portfolio in the key areas Inflammasomes & Innate Immunity, B & T Cells Immune Regulation, Immune Checkpoint Regulation, Neuroinflammation, Obesity/Metabolic Diseases and Bacterial and Viral Infection. AdipoGen Life Sciences is the manufacturer of many unique and high quality biochemicals and partners closely with Chemodex.
Natural Products / Rare Antibiotics
AdipoGen Life Sciences provides key standard research antibiotics, rare metabolites/antibiotics with new chemical structures (often only described once in literature and afterwards lost to research) or old already forgotten substances for lead drug research, seeking new “antibiotics” with new mode of actions and new molecular targets. In addition, these substances can be used for in vitro or in vivo studies based on their biological activities or as standard secondary metabolites, important as chemo-taxonomic markers of microbial species.

Inflammasomes & Innate Immunity
AdipoGen Life Sciences is an expert in inflammasome research. Inflammasomes are multi-protein complexes whose activity has been implicated in physiological and pathological inflammation.
The hallmarks of inflammasome activation are the secretion of the mature forms of caspase-1 and interleukin-1β (IL-1β) from cells of the innate immune system.
Beneficial inflammatory responses arise by maintaining homeostatic tissue function (detection and repair of tissue damages after trauma or pathogen invasion). Among the harmful inflammatory responses are particle-induced sterile inflammation, caused by host-derived particles such as monosodium urate (MSU) crystals, as well as environmental and industrial particles such as asbestos, silica and metallic nanoparticles.
Accumulating evidence also implicates inflammasome activity in numerous other diseases, including cancer and the development of metabolic diseases (like type 2 diabetes, atherosclerosis), some neurodegenerative diseases (like Alzheimer's, Prion, Parkinson's), autoimmune diseases (such as multiple sclerosis) and inflammatory bowel diseases.
AdipoGen provides a large selection of research reagents targeted to the various components of the inflammasome, thanks to a long-term collaboration with the Professor Jurg Tschopp group – the team who discovered the inflammasome in 2002.

AdipoGen also provides a large selection of research reagents targeted to the various components of the immune system. The innate immune system plays an essential role in the host's first line of defence against microbial invasion and involves the recognition of pathogen-associated molecular patterns (PAMPs) or endogenous danger signals through the sensing of danger-associated molecular patterns (DAMPs) by pattern recognition receptors (PRRs).

B & T Cells Immune Regulation / Immune Checkpoints / Immuno-oncology
TNF Superfamily – Key Cytokines in B & T cell Immuno-Regulation
TNFSF/TNFRSF together with the CD28-B7 family are two major co-signal pathways in T cell activation. CD28, ICOS, GITR, 4-1BB and OX40 pathways co-stimulate T cell activation. CTLA-4, PD-1, BTLA and CD160 pathways co-inhibit T cell activation.

Immune Checkpoint Reagents: Focus on T Cells
Cancer immunotherapy (also called immuno-oncology) is the artificial stimulation of the immune system to treat cancer, improving the system’s natural ability to fight cancer. Immune checkpoints are regulators of the immune system. These pathways are crucial for self-tolerance, which prevents the immune system from attacking cells indiscriminately. Inhibitory checkpoint molecules, such as CTLA-4, PD-1, PD-L1 or LAG-3 are targets for cancer immunotherapy due to their potential for use in multiple types of cancers. AdipoGen Life Sciences offers a broad panel of immune checkpoint reagents, including recombinant proteins, antibodies, and ELISA Kits.
